Materials
In total, 10% KOH, distilled H2O, 50% ethanol (EtOH, C2H6O), 75% ethanol, 95%
ethanol, acetic acid, clove oil, Canada balsam, xylene, microspatula, Minutin pin tool,
microscissors,syringe,Wilkey’sdoubleortriplestain,smallPetridishes(approximately
2 inches), hotplate, watch glass, slides and cover slips, slide oven or slide warmer and
slidelabelswereused.
Puparia
1)Placespecimensincold10%KOHfor12?24h,oriftheyaretobequicklyprocessed,
place them in a watchglass with KOH, and gently heat them on a hotplate for
approximately15 min. The temperature should not be high enough to allow KOH to
boil.
2)Remove the specimens from KOH, and place them in distilled water or acid alcohol
(50%ethanol:aceticacid4:1)toremoveorneutralizethealkalifor10?15min.
3a)Forpalepuparia:Add2dropsofdoublestainortriplestaintodistilledwateroracid
alcohol,andallowtositfor15min.
3b) For black puparia: Place the specimens in distilled water, and add 2 drops of H2O2
while observing the usually rapid bleaching. When black puparia have turned brown
andtranslucent,immediatelyproceedto4).
