Abstract :
As the rules of cattle and beef traceability were implemented in December 22th 2008
by ministry for food, agriculture, forestry and fisheries(MIFFF), we checked and
compared microsatellite(MS) markers and single nucleotide polymorphism(SNP) marker
method for the actual beef origin of Hanwoo, Korean cattle(Holstein) and imported
beef using from big market in Seoul city and Geynggi province. A total of 196 sample
including 90 samples with DNA over 50ng/ul concentration(50 samples of Hanwoo, and
40 imported beef) were collected from 15 big markets in Seoul city and Gyenggi
province from 2010 to 2011. We used polymerase chain reaction(PCR) of 45 Discerned
MS markers after extracting DNA from all beef samples, which method was already
developed by national livestock research institute. The PCR results were analyzed by
Applied biosystem 3130xl Genetic Analyzer(ABI, USA) and a statistics about individual
identification of types was estimated the individual discriminating power using
Geneclass 2 program(INRA, France) which is implied Bayesian methods. In addition we
used bead 90 single marker designed by SNP Genetics's KoBreed kit on same samples.
Identification of Hanwoo was recognized the more than 50% in the individual
discriminating power with MS method. The monitoring results showed that 71.4%,
9.7%, 17.3%, and 1.5% were positive with>95%, 85-94%, 50-84%,
samples, respectively. On the other hand, SNP method were decided that
point-estimate using the Genomstudio software in case of less than the 0.45 was
Hanwoo, more than 0.45 was non-Hanwoo. In this study, MS method were showed
the same result with SNP method on 90 beef These results suggest that beef labeling
system in big markets is properly conducted, and MS and SNP marker methods are
useful for traceability of Hanwoo, Korean cattle and imported breed.
